Sample Information

The Wales Cancer Bank collects frozen and paraffin embedded tumour and normal tissue, EDTA blood and serum from patients and EDTA blood from a non-blood related control (spouse, partner or friend). Samples are frozen in dry-ice cooled isopentane or liquid nitrogen and stored in freezers at -80 o C.

All frozen tissue samples have the presence of tumour verified by a pathologist and a digital image of the H & E slide is available for review.

DNA and RNA can be extracted from tissue, DNA from blood and Tissue Microarrays can be made on request.

Frozen blocks of tissue are sectioned in a cryostat and alternate sections used for separate extraction of RNA and DNA. The method ensures extraction of high quality DNA for use in techniques such as array CGH and high quality RNA for use in expression microarray systems.

DNA is extracted from tissue from 30 micron sections using a Qiagen DNA kit, and RNA using a Qiagen kit or Trizol method. Nucleic acids will be routinely supplied as 1 microgram in 50 microlitres for DNA and 5 micrograms in 50 microlitres for RNA.

RNA quality is assessed by 260/230 and 260/280 ratio using a nanospectrophotometer and then subjected to QA by Agilent Bioanalyser. Additional QA will be provided by RT-PCR for different sizes of amplimers generated from a housekeeping gene eg. PBGD. DNA quality is assessed by spectrophotometry and by 1% agarose gel electrophoresis.

DNA will be extracted from blood using a Gentra. DNA will be routinely available in 1 microgramme aliquots.

Researchers requiring different aliquot sizes should clearly state the required amount in their application form.
